Administration

• Inject once weekly on the same day each week.
• May be administered with or without food.
• Subcutaneous use only (abdomen, thigh, or upper arm).
• Rotate injection sites.
• Use a compatible disposable injection pen.
• Inspect solution; do not use if cloudy or discoloured.
• Allow to reach room temperature before injection.
• After first use, store below 30°C for up to 56 days.

Missed Dose

• If remembered within 5 days, administer as soon as possible.
• If more than 5 days have passed, skip the missed dose and continue the next scheduled dose.
• Do not administer two doses within 48 hours.

Storage

Before First Use

• Refrigerate at 2–8°C.
• Do not freeze.

After First Use

• Store below 30°C.
• Use within 56 days.

Common Side Effects

These effects are most common during dose escalation and usually improve with time:

• Nausea
• Vomiting
• Diarrhoea
• Constipation
• Abdominal discomfort

Indications

• Chronic weight management in adults with obesity (BMI ≥30 kg/m²).
• Weight management in overweight adults (BMI ≥27 kg/m²) with weight-related comorbidities.
• Adjunct to a reduced-calorie diet and increased physical activity.

Warnings & Precautions

• Gastrointestinal effects (nausea, vomiting, diarrhoea) are common during dose escalation.
• Risk of pancreatitis; discontinue if suspected.
• Increased risk of hypoglycaemia when used with insulin or sulfonylureas.
• May delay gastric emptying, especially with dehydration.

Contraindications

• Hypersensitivity to semaglutide, cagrilintide, or any excipients.
• Personal or family history of medullary thyroid carcinoma (MTC).
• Multiple Endocrine Neoplasia Syndrome Type 2 (MEN 2).

Special Populations

Elderly

• No dose adjustment required.
• Monitor renal function.

Renal/Hepatic Impairment

• No adjustment required for mild to moderate impairment.
• Use caution in severe cases.

Paediatric

• Safety and efficacy not established.

Pregnancy & Lactation

• Not recommended during pregnancy.
• Discontinue at least 2 months before a planned pregnancy.
• Unknown if excreted in breast milk; avoid use during breastfeeding.

Interactions

• May delay gastric emptying and affect oral drug absorption.
• Increased hypoglycaemia risk with insulin and insulin secretagogues.

Overdose

Symptoms:

• Severe gastrointestinal effects.
• Hypoglycaemia.

Management:

• Supportive treatment.
• Monitor glucose levels.

Disposal

Dispose of cartridges and needles in a sharps container according to local regulations.
